Determination of soluble and insoluble dietary fibres by en2ymabc-gravimetnc method applies to functional foods and establishes method for determining the total mass fraction of soluble and insoluble dietary fibers, including hemicelluloses, cellulose, pectins, resistant starches, lignin, etc. Range of measurement of the mass fraction of dietary fiber from 1% to 30% of the fat-free mass substances. The method is based on the enzymatic hydrolysis of starch and non-starch compounds with using a-amylase, protease and amyloglucosidase to mono-, di-, oligosaccharides and peptides. Dietary fibers are precipitated with ethyl alcohol, dried and determined gravimetrically. The total mass fraction of dietary fiber is expressed as a percentage (g/100 g). If necessary, the sample is ground in a homogenizer or laboratory mill. If the content fat in the sample exceeds 5%, it is extracted with petroleum ether at the rate of 25 cm3 per 1 g of the sample. The defatted sample is dried in an oven for one hour at a temperature 70°C and placed in a desiccator. When preparing and conducting the test, the following conditions must be met: (a) ambient temperature from 10 °С to 25 °С inclusive; and (b) relative air humidity from 40% to 70% inclusive.
